Study on Inhibition of Chorioepithelioma Cells' Growth by Laminarin Sulfate with Down-regulating the Expression of Heparanase

Objective: By viewing the effect of laminarin Sulfate (HPA inhibitors) to inhibit the growth of Chorioepithelioma cell JGE3, we object to interpret its mechanics and significance , find some new therapy drugs.Methods: by adding different concentrations of LS to the medium of choriocarcinoma cell, after 6h,we observe the cell growth state with the inverted microscope. RT-PCR was given to compare the expression of HPA in the groups. MTT colorimetric assay was used to detect cell viability. Flow cytometry (FCM) was used to analyze cell apoptosis.Results: (1)In those groups, with enhancing the dose of laminarin Sulfate (0.2ug/ml,0.4ug/ml, 0.6ug/ml, 0.8ug/ml),The effect of inhibition are significantly increased. The ratio of inhibition in four groups show24.62%、46.89%、62.58%、69.35% respectively, and the ratio of apoptosis are also increased, they are 7.53%、13.35%、21.46%、35.8% respectively.(2) In those groups , with enhancing the dose of laminarin Sulfate, the expression of Heparanase mRNA are gradually reduced.Analyzed by using correlative statistical test ,all the above results have obvious statistical significance, P < 0. 05.Conclusion: From the above results , we can get conclusion as below: laminarin Sulfate performs an significant effect on the proliferation of cancer cell. laminarin Sulfate inhibits the proliferation of cancer cell by regulating the expression of Heparanase.
